Characteristic Components Analysis in Lonicerae Japonica Flos by LC-UV

Lonicera Japonica Flos is a Traditional Chinese Medicine (TCM) used for the treatment of inflammation and infectious diseases. Chinese Pharmacopoeia has developed a method to support quality control efforts which includes the analysis of seven components in Lonicera Japonica Flos samples.

In this application brief, multiple marker compounds, including chlorogenic acid (CA), based on Chinese Pharmacopoeia standards, were analyzed using a PerkinElmer LC 300 UHPLC system, PerkinElmer Epic C18 column, a variety of chromatography consumables, and SimplicityChrom software.

Chlorogenic Acid Analysis in Children Throat-Reliever Granule by LC-UV

Utilized for throat inflammation in children, Xiaoer Yanbian Keli granules are a traditional Chinese formulated medicine, comprised of the Lonicera Japonica herbal plant. Chinese Pharmacopoeia developed standards in 2020 to ensure quality control, and has finalized the assay of a method in which the specification of the assay of chlorogenic acid is not less than 3 mg per bag.

In this application brief, quantification of the marker compound chlorogenic acid (CA) in a formulated drug, based on Chinese Pharmacopoeia 2020, is presented using a PerkinElmer LC 300 UHPLC with SimplicityChrom software, PerkinElmer Epic C18 column, and additional chromatography consumables.

HPLC Analysis of Acetaminophen, Aspirin and Caffeine Using an Epic C18 Column in Accordance with the USP

Acetaminophen, aspirin and caffeine used in combination are an effective method of treating acute headaches and migraines. The combination of these substances is considered superior to acetaminophen on its own when treating headaches. There are other benefits due to the individual components of the tablets. Acetaminophen (also known as paracetamol) is the most common analgesic and antipyretic around the world, it is an effective treatment for acute primary headaches on its own and can be combined with tramadol to treat cluster headaches. It has a very low risk of causing allergic reactions and so can be used by those with bronchial asthma. Aspirin is a first-line therapy to treat moderate to severe primary headaches as well as reducing pain, fever and inflammation. However, it is not taken by children under twelve due to the risk of Reye syndrome. Caffeine is considered an effective treatment for various types of headaches including post-dural puncture headaches and hypnic headaches. It also provides an increase in high energy exercise tolerance and reduces fatigue. These drugs combined are commonly sold under the brand names Anadin Extra in the UK and Excedrin in the U.S. In 2019 approximately 13.1 million units of Excedrin Migraine were sold in the United States. This application brief describes the use of an ES Industries Epic C18 column for the analysis of acetaminophen, aspirin and caffeine in accordance with the official Acetaminophen, Aspirin and Caffeine Tablets United States Pharmacopeia (USP) method for the assay.

HPLC Analysis of Amlodipine Besylate in Accordance with the USP

Amlodipine besylate belongs to a class of drugs known as dihydropyridine calcium channel blockers. It is primarily used to treat hypertension and angina as well as other conditions which arise as a result of coronary artery disease. As a calcium channel blocker, it prevents diffusion of calcium into cardiac muscle and the smooth muscle of the blood vessels. This causes vasodilation, lower heart contractility and therefore lower blood pressure. In addition, production of nitric oxide (a known vasodilator) increases. Amlodipine is listed on the World Health Organization’s (WHO) list of essential medicines as an antihypertensive medicine, meaning that it considered among the safest and most effective compounds needed to meet the requirements of a basic healthcare system. This application brief describes the use of our Epic C18 column to analyze amlodipine besylate in accordance with the official USP monograph.

HPLC Analysis of Betamethasone Dipropionate Using Epic C18 HPLC Column

Glucocorticoid steroids work by suppressing various aspects of the human immune system in conditions where hyperactivity can cause poor health through allergies, inflammation, and autoimmune dysfunction. Betamethasone dipropionate belongs to this class of steroids. It is used for its high potency as an anti-inflammatory and immunosuppressant in the treatment of diseases such as eczema, dermatitis, and psoriasis. Betamethasone dipropionate is classified as a ‘super-potent’ steroid in the treatment of psoriasis in comparison with betamethasone valerate (another common analogue of betamethasone) which is rated as upper mid-strength. This application brief describes the use of an Epic C18 column for the analysis of betamethasone dipropionate in accordance with the official USP monograph.

HPLC Analysis of Common Ginsenosides Found in Ginseng Using the Chinese Pharmacopeia

The term ginseng can refer to several natural products and herbal remedies usually produced from the plant, panax ginseng. Panax ginseng is considered a traditional Chinese medicine (TCM) as it has a reported wide range of medicinal properties such as antioxidant effects and promotion of immune function.1 The chemistry of ginseng is still not fully understood and more research is required to understand its functions as it becomes increasingly popular in western culture. The main active components of ginseng are ginsenosides of which there are over 100 in various forms of panax ginseng. Ginsenosides are named as Ginsenoside Rx, with x representing the movement on a thin layer chromatography plate, x represents a letter from a-h with a being the most polar and h being the least. This application brief describes the use of an Epic C18 to analyze three ginsenosides using a method based on the Chinese Pharmacopeia (ChP).

HPLC Analysis of Finasteride Using an Epic C18 HPLC Column

Finasteride is an antiandrogenic compound or ‘testosterone blocker’ which is used as a treatment for an enlarged prostate and can treat male hair loss. It inhibits the production of the enzyme Type-II 5a-reductase which converts testosterone into the more active metabolite 5a-dihydrotestosterone (DHT). DHT is responsible for the development and enlargement of the prostate and can contribute to hair loss by shortening hair follicles and causing them to fall out more quickly. By suppressing production of DHT, finasteride directly targets the molecule responsible for stimulation of prostatic growth. Finasteride is commonly sold under the brand name Proscar and in 2016 was the 75th most prescribed drug in the USA. This application brief describes the use of an Epic C18 column to analyze finasteride in accordance with the official Finasteride Tablets USP monograph.

Oleanlic Acid and Ursonic Acid Analysis of Chaenomelis Fructus by LC-UV

Chaenomeles, the fruit of the Chinese flowering quince, is utilized in Traditional Chinese Medicine (TCM) for its analgesic, anti-inflammatory, antispasmodic and astringent properties, and has also been shown to aid in digestion. For implementation of quality control processes, Chinese Pharmacopoeia has released the assay of oleanolic acid and ursonic acid of the herbal plant. In this work, the analysis of these two compounds, following Chinese Pharmacopoeia 2020 standards, is presented utilizing a PerkinElmer LC 300 system with SimplicityChrom software, PerkinElmer Epic C18 column and additional chromatography consumables.

Analysis of 16 Cannabinoids Using the PerkinElmer LC 300 HPLC System with PDA Detection

Accurate identification and quantification of cannabinoid content is a critical step in the process to bringing safe and high quality cannabis flower and fortified products to market. As the legalization of recreational cannabis use continues throughout the world, fast and efficient liquid chromatography methods for the determination of cannabinoid content are need to satisfy both regulatory and consumer demands.

In this application note, a fast and simple method for the determination of 16 common cannabinoids is presented. Utilizing a PerkinElmer LC 300 HPLC system with a PDA detector, the method results in the elution of all sixteen compounds in under seven minutes, with LOQs = 0.15 µg/mL for most analytes.

Analysis of Common Preservatives in Personal Care Products by HPLC with UV Detection

Parabens are utilized as an antimicrobial preservative in many commercial offerings including cosmetics, personal care products and foods, and offer both chemical and thermal stability to products. Although parabens increase the safety of products, recent studies have suggested that parabens may exhibit weak endocrine disrupting effects on users of the products, an effect that can be further intensified by the ability of parabens to be absorbed by the skin without being broken done by esterase enzymes.

In this application note, a fast, robust and sensitive method utilizing a PerkinElmer LC 300 HPLC system with Multi-Wavelength UV/Vis (MWD) detector for the analysis of 2-phenoxyethanol and seven commonly used parabens in eight different commercially available cosmetic and personal care products is presented.

Analysis of Phytoestrogen Isoflavones in Dietary Supplements by HPLC/UV

Isoflavones are water-soluble compounds found in many plant and food sources. Designated as phytoestrogens, isoflavones are structurally similar to to the female hormone estrogen, and have been shown to exhibit a number of beneficial impacts to human health, such as a reduction in the development of certain cancers, as well as an improvement in cholesterol ratios. Studies also suggest, however, that the phytoestrogenic nature of isoflavones can also lead to an exacerbation of existing thyroid disorders, owing to a disruption to the endocrine system. As such, precise quantification of isoflavone content in nutraceutical products is paramount to ensure label-claim accuracy and consumer safety.

In this application note, an efficient and reliable method for the determination of six common isoflavone compounds is presented. The method, which was developed in accordance with the USP monograph for soy isoflavones in dietary supplement capsules, utilizes a PerkinElmer LC 300 UHPLC system, with subsequent detection of compounds achieved utilizing an LC 300 PDA detector.

Analysis of Saccharides in High Fructose Corn Syrup

High fructose corn syrup (HFCS) as a substitute for sucrose in a wide variety of products, including baked goods, sauces, beverages, canned fruits and dairy products. HFCS is produced from regular corn syrup, with an enzymatic process that isomerizes some of the corn syrup's glucose into fructose. The fructose concentration can vary, depending upon the intended use, and is commonly produced at concentrations of 42% (HFCS-42), 55% (HFCS-55) and 90% (HFCS-90).

The US Food and Drug Administration Code of Federal Regulations have incorporated identity standards for the sugar composition of HFCS, specifically around fructose content where HFCS-42 cannot be less than 42% fructose and HFCS-55 cannot be less than 55% fructose. This application note demonstrates the suitability of the PerkinElmer LC 300 HPLC system for performing saccharide content determination via method CRA SACCH.03 through the analysis of nine commercially available high fructose corn syrups.

Analysis of Sugars in Maple Syrup by HILIC with Refractive Index Detection

Maple syrup, a common natural sweetener, is consumed regularly by households around the world. Quantification of sugar content in maple syrup is paramount to satisfy product labeling requirements and ensure label-claim accuracy. Although sucrose is the primary sugar component in maple syrup, a complete identification and quantification of the other complex carbohydrates, including glucose, fructose and maltose, is essential.

In this application note, a robust and efficient method for the determination of the four aforementioned complex carbohydrates is presented. Utilizing an LC 300 HPLC system, a hydrophilic interaction chromatography (HILIC) technique is utilized with subsequent detection achieved utilizing an LC 300 Refractive Index (RI) detector.

HPLC Analysis of Lamotrigine Using an Epic C18 SD Column in Accordance with the United States Pharmacopeia

Lamotrigine is an antiepileptic drug in the phenyl triazine class used in the treatment of both epilepsy and as a mood stabilizer for bipolar disorder. It is the first medication since lithium to be granted US FDA approval for maintenance treatment of bipolar type 1, and is approved for use in more than 30 countries.

In this work, the analysis of Lamotrigine, in accordance with the USP monograph, is detailed. Utilizing a PerkinElmer LC 300 liquid chromatography system with SimplicityChrom™ CDS and PerkinElmer Epic™ C18 SD column, the analysis is efficient and accurate, with tailing factor and repeatability results achieved are well within the monograph acceptance criteria.

Paeoniflorin Analysis in Paeoniae Radix Alba by LC-UV

Paeoniae radix alba, also known by its Chinese phonetic name as Baishao, is a Traditional Chinese Medicine herbal plant used to treat inflammatory disorders. Paeoniflorin, the major active component extracted from Paeoniae radix alba, offers proven efficacy against gynecological disorders, pain, and hyperactivity of liver-yang, among other disorders.

Technical Note

A Comparison of Fully Porous C18 Reversed Phase HPLC Columns

The most commonly used LC reversed-phase alkyl bonded stationary phase is octadecyl carbon chain (C18)-bonded silica, which is denoted as USP classification L1. C18 columns have a broad applicability from pharmaceuticals to food and environmental analyses. However, not all C18 columns are alike. Simply swapping a C18 column from one manufacturer to another can result in differences in retention time, resolution and even selectivity. Differences can arise due to variations in hydrophobicity, silanol activity, packing quality, particle size distribution, and silica purity, to name a few.

This technical note provides details of a comparative study between twenty-four silica-based C18 phases, from a number of manufacturers covering the following areas:

Clone Columns Aviator C18 and ACE C18

Legacy methods, by their very nature, often use older column technologies. These older phases can be accompanied by larger variations in batch-to-batch performance which can lead to inconsistent results and cause out of specification (OOS) occurrences. With routine analysis, often completed with compliant procedures, any unplanned downtime to investigate OOS instances can impact productivity. Our range of clone phases offer a cost-effective comparable alternative to many of the older leading brands, whilst ensuring consistency and stability in analysis. Better lot-to-lot reproducibility is also achieved due to more stable production methods especially when compared to older brands. Our Aviator™ line exhibits similar selectivity to Avantor ACE® HPLC columns. Aviator C18 provides a rugged, reproducible starting point for method development and applications with analytes differing in hydrophobicity, polar, moderately polar and non-polar analytes, uncharged acids and bases, and ionized acids or bases using ion-pairing. Other Aviator phases are also available including AQ, C8, C4, Cyano and Phenyl. Other Harmony phases are also available including C8 and C4.

The injection of an accurate and reproducible sample volume into a UHPLC system is challenging, but essential to ensuring a high quality analysis. The patented PerkinElmer Intermediate Loop Decompression (ILD) valve, featured in the LC 300 Autosampler, is a 7-port valve with a strategically placed radial groove in the rotor seal which ensures the injection of an accurate and reproducible sample volume, while also increasing the lifetime of columns used in analysis.
