N-Hydroxysuccinimide (NHS)-ester labeling reagents, such as VivoTag®-S reagents, are commonly used for labeling proteins. The NHS ester moiety reacts with amino groups at pH 7-9 to form stable amide bonds. Lysines within proteins, including antibodies, are available as targets for this chemical conjugation.,Protein labeling efficiency may vary depending upon the type of protein labeled, so different conjugation ratios may need to be attempted to attain success. For Near Infrared (NIR) fluorophore conjugation, ratios of fluorophore to protein of 4:1 have been seen to generate effective imaging agents. Not all antibodies or proteins (independent of target specificity) make good imaging agents, due to long half-lives and/or excessive accumulation in non-target sites.
Researchers trust our in vivo imaging solutions to give them reliable, calibrated data that reveals pathway characterization and therapeutic efficacies for a broad range of indications. Our reagents, instruments, and applications support have helped hundreds of research projects over the years. And our hard-earned expertise makes us a trusted provider of pre-clinical imaging solutions— with more than 9,000 peer reviewed articles as proof.
We have developed a novel FAST (“Fluorescent Activatable Sensor Technology”) in vivo imaging agent based on an angiotensinogen substrate sequences that is selectively cleaved and activated by rodent renin. ReninSense680 FAST, a PKM-modified agent suitable for invivo administration, distributes to the kidney, is activated in a model of renin upregulation, and provides a tool for in vivo imaging and quantification of the changes in renin activity inkidneys and plasma.
VivoTag-S® 680 is an amine reactive near-infrared (NIR) fluorochrome (NHS ester) for labeling biomolecules for in vivo imaging applications.
The primary goal of preclinical imaging is to improve the odds of clinical success and reduce drug discovery and development time and costs. Advances in non-invasive in vivo imaging techniques have raised the use of animal models in drug discovery and development to a new level by enabling quick and efficient drug screening and evaluation. Read this White Paper to learn how preclinical in vivo imaging helps to ensure that smart choices are made by providing Go/No-Go decisions and de-risking drug candidates early on, significantly reducing time to the clinic and lowering costs all while maximizing biological understanding.
