The VICTOR Nivo system is a high-performance filter-based multimode microplate reader equipped with all major detection technologies:

It’s a compact, light-weight multimode plate reader designed for life science research laboratories performing routine low-throughput assays, or assay development work, with diverse application requirements. When combined with the stacker, throughput for the VICTOR Nivo is increased and productivity is enhanced by enabling users to set up walk away automation so they can focus on their research and not on managing plates.

The VICTOR NIVO system’s software has a modern, workflow-oriented user interface which is easy to learn and use and includes pre-written application protocols to get users productive quickly. In addition, MyAssays Desktop Standard software is provided for data analysis. With our Enhanced Security software option, it contains technological tools to help guide you through the complexity of compliance for 21 CFR Part 11.

The browser-based control software is independent of operating system, so it can be controlled via a variety of touchscreen devices, for example Windows or Mac OS PCs or tablets.

The VICTOR Nivo microplate reader can be operated via WiFi or a network connection, enabling experiments to be set-up, run and monitored remotely, such as from outside a biosafety cabinet or from a neighbouring lab.

Dynamic Rainbow Filter Exchanger

Every VICTOR Nivo comes rainbow-ready with our dynamic double-layered filter wheel system that can store up to 32 filters. When fully loaded, the system is equipped to address virtually any application, with better sensitivity than a monochromator.

Filters are:

  • Exchanged between the inner and outer wheels
  • Used for either excitation or emission light path
  • Locked in position so they don't get lost - ideal for multiuser labs

Detection Technologies

For absorbance measurements using the VICTOR Nivo multimode plate reader, there is a choice of either a filter- or a spectrometer-based system. With the spectrometer-based plate reader, full spectrum absorbance measurements are ultra-fast – 230 to 1000 nm at selectable resolutions (2.0 nm, 5.0 nm, 10 nm) in less than 1 second per well.

Up to eight discrete wavelengths can be detected at once in a single measurement with no wavelength switching, and the spectrometer-based VICTOR Nivo system also allows the detection of a wide range of dyes or measurement of samples with unknown absorbance spectra.

Adaptation of a Bacterial Growth Detection Assay on the VICTOR Nivo for Measurement of Antibiotic Effects

Monitoring of bacterial growth is a fundamental technique in microbiology. The most widely used method for enumerating bacteria is plate-based counting, which requires serial dilutions of the bacterial stock culture and manual counting of colonies after plating. The procedure is very time consuming and cost intensive, as plates must be prepared, inoculated and counted manually. Running the assay in a microplate format and using a plate reader to measure bacterial growth greatly reduces consumables and waste. Automating sample preparation with an automated liquid handler helps to further reduce hands-on time and improve precision.

In this application note, we present the adaptation of a microplate-based assay on the VICTOR® Nivo multimode plate reader for automated detection of bacterial growth assay, using the readout for optical density (OD)/turbidity. The integrated temperature control and shaking function of VICTOR Nivo allows the realization of controlled assay conditions, helping to prevent common issues such as condensation under the plate lid, and allowing continuous detection of changes in turbidity over time. High-Throughput DNA Quantification for Next Generation Sequencing on the VICTOR Nivo

There are many different methods of quantification available on the market for DNA samples and each can vary in precision, accuracy and throughput. Typically, DNA in next generation sequencing (NGS) workflows is measured using tube-based fluorescent assays such as Qubit® or qPCR. However, since samples have to be measured one at a time, this can create bottlenecks in a high-throughput lab. Using an automated liquid handler and plate reader instead of manually pipetting tube-based measurements can significantly decrease measurement time and increase throughput.

Quantification of ADAMTS13 Activity by FRET Assay and Kinetic Data Analysis Using MyAssays Desktop on the VICTOR Nivo

In this application note, the VICTOR® Nivo multimode plate reader is used in combination with MyAssays® Desktop Software to perform kinetic assays and analyze data in a straightforward and efficient manner.

ADAMTS13 is a biomarker of thrombotic thrombocytopenic purpura (TTP), a rare disease that can be controlled by plasma exchange which is supposed to supply ADAMTS13 to patients and/or to remove neutralizing autoantibodies against ADAMTS13.

VICTOR Nivo was used to measure the activity of the enzyme ADAMTS13 in plasma samples, using the commercial substrate “FRETS-VWF73”. As variant of the original protocol, PerkinElmer half-area plates were used to save reagent and sample (assay volumes reduced by half as compared to the original protocol).

Quantification of Proteins by UV-Vis Absorbance Using VICTOR Nivo with Microvolume Plates

Knowing the protein concentration is commonly required in studies of protein biochemistry and molecular biology. A direct method of measuring protein concentration is to determine the absorbance spectrum at 280 nm. With this UV based approach, it enables scientists to rapidly determine the concentration of protein with a procedure that is simple and nondestructive, does not need special reagents or standard curves, and consumes very little sample.

DNA Quantification Using the VICTOR Nivo and Micro-Volume Plates

All life science related fields from basic research in academia to commercial diagnostics laboratories rely on quick and simple, yet highly accurate quantification of nucleic acids. Commonly, photometers are employed to determine quality, purity, and concentration by measuring the absorption of the probe using light in the ultra-violet (UV) range with wavelengths of 230, 260, 280, and 320 nm. While the absorption of 230 nm light indicates contamination with organic compounds, salt, sugar, or phenol, 320 nm serves as background correction. Nucleic acids and proteins show absorption maxima at 260 and 280 nm, respectively.

Using the VICTOR Nivo for Cell-Based HTRF Assays

Cell-based assays are routinely employed in R&D to reveal compound induced effects in vitro. In this technical note, we describe the usage of the VICTOR® Nivo multimode plate reader for alpha-tubulin acetyl-K40 detection in a cellular assay based on HTRF® technology. Tubulin acetylation is widely studied due to its dysregulation in human neurological disorders. To demonstrate the HTRF assay principle on the VICTOR Nivo, we used mouse fibroblasts (NIH-3T3 cells).

Ensuring that laboratory processes are compliant with the regulations of the U.S. Food and Drug Administration (FDA), such as 21 CFR Part 11, and other references, e.g. Annex 11 published by the European Commission, can be time-consuming and laborious, requiring meticulous documentation of procedures and record-keeping.
