Metalloproteinase Inhibitor 1 / Tissue Inhibitor of Metalloproteinases (TIMPs) are a family of inhibitors binding to most matrix metalloproteinases (MMPs) through their interaction with the Zn2+ binding pocket of active MMPs. They are also inhibitors of proteins from the ADAM and ADAMTS families. TIMP1 is a glycoprotein of 184 amino acids in its active form. High levels of TIMP1 were observed in many types of cancer, including breast and colon cancer. Increasing the expression of TIMP1 suppresses the invasiveness of some cancers like melanoma and fibrosarcoma cells. Many studies demonstrated that TIMP1 can be used as a prognostic marker of survival in patients with breast and colorectal cancer.
AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
