Human Trefoil Factor 3 (TFF3) is a small 6.7 kDa homodimeric protein. It is part of a small family of secreted proteins characterized by at least one copy of the trefoil motif, a 40 amino acid domain containing three conserved disulfide bonds. TFF3 is normally produced by goblet cells of the intestine and the colon, but expression was also observed in other tissues. The expression in the gastrointestinal mucosa seems to play an important role in its self-protection, through stabilization of the mucus layer, promotion of epithelial cell migration, and overall healing and repair of the intestinal mucosa. Overexpression of TFF3 has been associated with many forms of cancer, including gastric, prostate, pancreas, and breast cancers. It is being considered as a serum biomarker for endometrial carcinomas.
AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
