Mucin-16 / Ovarian Carcinoma Antigen CA125 (CA125) is a 22 152 amino acid transmembrane protein with a small intracellular segment and a large extracellular domain. Cleavage, induced by intracellular phosphorylation, causes release of the heavily glycosylated extracellular domain. Association of several molecules of the secreted form are known to occur through interchain disulfide bridges creating a large gel-like matrix that may function as a protective, lubricating barrier against infection at mucosal surfaces. It is best known as a marker for ovarian cancer, but it may also be elevated in other cancers including those originating in the endometrium, fallopian tubes, lungs, breast, and gastrointestinal tract. CA125 levels may also be elevated in a number of relatively benign conditions such as endometriosis, several diseases of the ovary, pregnancy, and inflammatory diseases in the abdominal area.
AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
