DAP12, also known as TYRO protein tyrosine kinase-binding protein, is a signaling adaptor for several cell surface receptors of the innate immune system. Crosslinking receptors associated with DAP12 leads to phosphorylation of tyrosine residues in their cytoplasmic ITAM by SRC family kinases, and initiates downstream signaling through the activation of SYK or Zap-70 and additional downstream targets. The TREM2-DAP12 receptor complex appears to play a continuous surveying role by scanning the neuronal microenvironment and maintaining a healthy brain homeostasis in Microglia cells.
