ELISA, western blot, and other immunodetection technologies have commonly been used for quantitation of biomarkers and other analytes in a variety of sample types. These methods necessitate long, tedious protocols and often provide limited detectable range.
LANCE® (Lanthanide Chelate Excite) TR-FRET is a homogeneous (no-wash) alternative to traditional ELISA. LANCE is a proximity assay that combines the benefits of time resolution (TR) with fluorescence resonance energy transfer (FRET) principles by using energy donor and energy acceptor fluorophore labels. It’s everything you love about TRF detection with faster time to results and no wash steps.
In a LANCE TR-FRET immunoassay, a target specific antibody is either conjugated directly to or is captured by an antibody conjugated to a Europium chelate. A second, target specific, antibody is either conjugated to or is captured by an antibody conjugated to the ULight™ acceptor dye. These two target- specific antibody capture the analyte, brining the Europium chelate donor and the ULight acceptor dye into proximity. Upon excitation at 320 nm or 340 nm, the Europium chelate is excited and energy is transferred to the ULight dye, which emits light at 665 and 615 nm. The amount of light emitted is proportional to the amount of analyte present in the sample.
