Aflatoxins are a group of molecules in the mycotoxin family. They are produced by Aspergillus flavus and Aspergillus parasiticus that are found on agricultural crops such as maize (corn), peanuts, cottonseed, and tree nuts. These molds thrive in warm humid environments. Aflatoxins are small organic compounds that survive digestion in both humans and animals. Chronic exposure to aflatoxin can lead to many liver disorders including hepatic necrosis, cirrhosis and carcinomas. The amount of aflatoxin present in food and animal feed is highly regulated by agencies around the world.
AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In this AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while anti-HRP is conjugated to AlphaLISA Acceptor beads. In the presence of an HRP-labeled aflatoxin tracer, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm. With increasing amounts of aflatoxin from sample, there is a decrease in signal.
