Renin (or angiotensinogenase) is a 44 kDa aspartyl protease that catalyzes the first step in the activation pathway of angiotensinogen. Prorenin is the precursor of renin. The majority of prorenin is secreted constitutively to the circulation while the rest is kept in secretory granules where it is converted to renin by specific proteases and then released upon demand. The final product of the renin-angiotensin system is angiotensin II, which is the main physiologically active substance involved in systemic blood pressure regulation and electrolyte homeostasis. Gene mutations in renin have been shown to cause familial hyperproreninemia. Renin serum levels are commonly measured for prognosis of cardiovascular disease.

AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
