N-Cadherin (NCAD or Cadherin-2) is a 99.7 kDa (906 AA) transmembrane protein composed of 5 extracellular cadherin repeats. It is expressed in a variety of tissues including cardiac and neural tissues, where its role is to mediate cell-cell interactions by interacting with cadherins on adjacent cells. N-Cadherin is commonly found at elevated levels in some cancer cells and due to its role in epithelial –mesenchymal transition it is thought to play a vital role in cancer metastasis. As such, N-cadherin has garnered attention as an important biomarker. This AlphaLISA kit has been designed to detect and quantify N-Cadherin in cell lysate and tissue homogenates.
AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
