In this AlphaLISA competition assay, biotinylated cAMP probe binds to Streptavidin-coated Alpha Donor beads, with anti-cAMP antibody conjugated to AlphaLISA Acceptor beads. In the absence of endogenous cAMP from the sample, the beads come into close proximity. The excitation of the Donor beads triggers the release of singlet oxygen molecules that diffuse to the Acceptor beads, resulting in a sharp peak of light emission at 615 nm. With increasing concentrations of endogenous cAMP from the sample, fewer Donor and Acceptor beads come together, resulting in a decrease in signal.
This assay kit contains enough reagents to run 500 wells in 96-well or 384-well format, using a 50 µL final reaction volume (20 µL of sample or cells).
