Four main classes of immunoglobulins have been recognized in the cow: IgG (comprising IgG1 and IgG2 subclasses), IgA, IgM, and IgD. Two forms of bovine IgA have been identified: a normal serum IgA with no secretory antigenic determinant and a small amount of secretory IgA in milk and colostrum. Secretory IgA most often appears as a dimer of the 4-chain IgA unit plus a glycoprotein called the "secretory" piece. Typically, bovine samples contain about 0.0 to 32.0 mg/mL of IgA depending on the type of body fluid (milk, colostrum, serum, plasma, saliva or tears) and whether the samples were collected pre-or post-partum. The present kit permits detection of bovine IgA (i.e. analyte) in bovine serum and plasma, as well as other biological fluids, such as milk and colostrum.

AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
