Alpha-fetoprotein (AFP) is a 67 kDa plasma glycoprotein, present in monomeric, dimeric and trimeric forms, produced by the yolk sac and the fetal liver during fetal life. Thought to be the fetal equivalent of serum albumin, it binds to estradiol, copper, nickel, fatty acids, and bilirubin. It serves as a carrier protein in the blood. AFP expression in adults is often associated with hepatoma or teratoma, as normal adult levels are low, but detectable. The level of AFP in amniotic fluid is used to measure renal loss of protein to screen for spina bifida and anencephaly.
AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
