Alpha2-Macroglobulin (a2-Macroglobulin) is a glycoprotein that circulates at a constant level in the blood as a tetramer composed of 4 identical subunits of about 180 kDa. a2-Macroglobulin is mainly secreted by the liver / hepatocytes, but also macrophages, fibroblasts, astrocytes, sertoli cells, adrenocortical cells and certain types of tumors. a2-Macroglobulin is a member of the macroglobulin protein family with similar function: inhibitors of all classes of endopeptidases. a2-Macroglobulin mediates the clearance of endopeptidases from the plasma, but in some cases can also act as a carrier protein in blood for interleukin-6. An abnormal increase of a2-Macroglobulin in blood has been suggested to be associated with multiple sclerosis, glomerular disease, and liver diseases.

AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
