Ratiometric imaging is the division of one channel by a second channel to derive a third, ratiometric channel. This technique is extremely attractive because it corrects for unequal dye loading (or sensor expression), dye leakage and photo-bleaching. Ratiometric is commonly used to monitor changes in intracellular calcium ions. Ratio imaging often requires high speed acquisition and consequently is usually performed only in 2D.
