Automated Normalization using the JANUS Workstation

Many genomic samples have variable DNA contents and require dilution to a constant amount of nucleic acid in order to successfully perform downstream analyses such as multiplexed PCR-based genotyping. The JANUS® Automated Workstation offers a complete genomics solution that includes a normalization and dilution procedure following DNA quantification. DNA concentrations can be measured using a fluorescent DNA stain or determination of the absorbance ratio 260/280 nm, or a luminescence assay.
After quantification, the JANUS DNA Normalization Protocol uses output DNA concentration data to ‘intelligently’ dilute the DNA to a desired target amount per sample well, usually 1 ng. Once the normalization protocol is completed, samples are ready for further analysis.
The JANUS automated liquid handling workstation can enable:
- Normalization and dilution using JANUS Normalization Protocol
- Normalization using JANUS Victor Normalization Option with Workout data analysis software package
- Normalization following fluorescent quantification using OliGreen® or PicoGreen® assay
- Normalization following luminescent quantification using human specific Promega AluQuant assay
