Quantification and Normalization using the JANUSAutomated Workstation
Accurate quantification and normalization of genomic DNA samples are critical for successful genotyping due to variable DNA properties and the need to create uniform concentrations. Reliable and accurate normalization is required to produce quality genotyping profiles, microarrays and DNA sequence data for molecular diagnostic and research laboratories. The
JANUS® Automated Workstation offers two different DNA quantification and normalization procedures: choose a plate reader to detect and measure UV absorbance/fluorescence OR a highly sensitive real-time quantitative PCR (RT-QPCR) method.
JANUS Automated Plate Reader Normalization
Our family of JANUS automated liquid handling workstations can easily integrate with any of our detection systems including the Victor3™ or Envision® instruments to support your normalization technique. DNA concentrations are measured using a fluorescent DNA stain or determination of the absorbance ratio 260/280 nm. After quantification, the JANUS Nucleic Acid Normalization Protocol fully automates the DNA dilution to your target concentration. Adding our software provides full walk-away automation for nucleic acid quantification and normalization in a 96- or 384-well format for one or multiple plates. Our user-friendly templates simplify sample quantification and normalization assay set up, as well as allow you to define parameters such as normalization concentration, final sample and final normalization volumes.
JANUS Automated RT-QPCR Normalization
With this technique, the JANUS automated liquid handling workstation is able to integrate with third-party thermal cyclers, to acquire the necessary date and automate the DNA dilution to your target concentration. In addition, the data is used to normalize pipetting steps over a 10,000-fold concentration range. After normalization is achieved, you can initiate additional protocols for downstream analysis procedures such as PCR setup or upstream QPCR assay setup.
