RNA Interference (RNAi)

RNA interference (RNAi) is an enabling technology in the field of functional genomics. It facilitates genome-scale loss-of-function screens in cultured cells and is ideally suited to be used for High Content Screening for target identification and signal transduction pathway analysis.

Usually a set of two to four siRNAs per gene are transfected into the cells. The double-stranded siRNAs are unwound and the antisense strand directs recognition of the target mRNA sequences. The mRNA, which is homologous to the introduced siRNA, is then subsequently degraded, resulting in a loss of gene function in the cell sample.
