AlphaScreen®SureFire® for Cell-based Kinase & Cellular Signaling Proteins
AlphaScreen
®SureFire® is a unique platform for measuring endogenous proteins in cell lysates, as well as key disease-related signal transduction pathways. It enables the highly sensitive and precise interrogation of various signaling pathways, receptors and kinase targets, and the measurement of full-length, endogenous protein phosphorylation in a cell-based format.AlphaScreen
®SureFire® makes it possible to culture cells and detect key analytes using an "All-in-One- Well" format. Alternatively, cells can be used in multiple assays analyzing pathways or allowing for kinase profiling. This eliminates time-consuming separation and wash steps.
AlphaScreen
®SureFire® kits are used in conjunction with AlphaScreen Protein A kits, which include the streptavidin-coated Alpha Donor bead and the Protein A-coated Acceptor bead.
Applications
Ideal for screening GPCRs and growth factor receptors, and for intracellular kinase inhibitors of MAPK and other signaling pathways.
Features/Benefits- Homogenous: No wash steps, miniaturizable for high throughput
- Non-radioactive: Eliminates special reagent handling and disposal, faster read out
- Flexibility: Use in any cell line, including primary cells
- Sensitivity: Can be used on cells over-expressing cloned targets or with cells expressing physiologically relevant endogenous levels of receptor target
- Automatable: Easy to automate with HTS-proven AlphaScreen technology
- Ultra HTS compatible: Readily adaptable to 96-, 384- and 1536-well formats
- Multiple assay options: Permits interrogation of various signaling pathways, receptors and kinase targets
